Cytocompatibility, cytotoxicity and genotoxicity analyses of dental implants JBAG
Reigosa,M.; Labarta,V.; Molinari,G.; Bernales,D..
Different materials are used for prostheses in dental medicine, though titanium is the most frequent. In the present report, several commercial dental implants and their potential capacity to integrate to bone tissues have been analysed by means of cytotoxicity, genotoxicity and cytocompatibility techniques. Cultures from osteosarcoma-derived UMR 106 cell line were used for bioassays since they showed several osteoblast properties. Samples of dental implants provided by B&M national company (ASTM titanium 8348 GR2, acid-etched surface) were compared with some others commercialized by foreign firms in Argentina (Nobel: anodic-oxidized surface, and 3i: sandblasted surface). Using the neutral red uptake test, lysosome and alkaline phosphatase enzymatic...

Cytological and cytogenic effects induced by thiabendazole on Allium cepa root meristem JBAG
Andrioli,Nancy B; Mudry,Marta D.
The toxicity and genotoxicity of thiabendazole (TBZ) in its commercial formulation Foldan® was studied in Allium cepa meristematic cells at concentrations ranging between 10 and 250 µg / ml. Our results show that exposure to TBZ induces a significant increase in the frequency of anaphase telophase chromosomal aberrations, micronuclei and binucleated cells at all the concentrations of TBZ assayed. The frequency of anaphase telophase aberrations was observed to reach a plateau at the concentration of 50 µg / ml of TBZ, which could be probably due to TBZ mitotic toxicity manifested at concentrations of 100 and 250 µg / ml. An increase was also observed in the rate of metaphase and anaphase, which was, in turn, indicative of alterations in chromatid...

Cytotoxic activity of BCG-activated macrophages against L929 tumor cells is nitric oxide-dependent BJMBR
Nascimento,F.R.F.; Ribeiro-Dias,F.; Russo,M..
The tumoricidal activity of activated macrophages has been attributed largely to the release of tumor necrosis factor (TNF), or to the production of reactive oxygen or nitrogen intermediates. The L929 tumor cell line (a murine fibroblast-like cell) when treated with actinomycin D (ActD) has been used to measure TNFa cytotoxicity. In the present study, we determined the cytotoxic activity of BCG-activated peritoneal macrophages against ActD-untreated L929 tumor cells. Furthermore, we measured the production of hydrogen peroxide (H2O2), nitric oxide (NO) and TNF by macrophages cultured in the presence or absence of L929 cells. As expected, BCG-activated macrophages produced significant amounts of H2O2 (16.0 ± 3.0 µM),...

Cytotoxic and antibacterial properties of Mytilus galloprovincialis, Ostrea edulis and Crassostrea gigas (bivalve molluscs) hemolymph ArchiMer
Hubert, Florence; Van Der Knaap, Wil; Noel, Thierry; Roch, Philippe.
Mussel (Mytilus galloprovincialis) plasma contains cytotoxic activity against both vertebrate (erythrocytes and mouse tumour) and protozoan cells. Procaryotes (Escherichia coli and Vibrio alginolyticus) were not sensitive to the cytotoxicity. The activity was still present in dialyzed samples but was inhibited by heating at 45°C. Large individual variability which was not correlated with protein concentration and an increasing number of reactive specimens following injection, suggested that naive mussels were in various stages of immune response. Purification by anion exchange chromatography followed by gel filtration revealed a 320 kDa cytotoxic polymeric protein that acts through a polymerization process after binding onto target cell membranes as...

Cytotoxic and pro-apoptotic action of MjTX-I, a phospholipase A2 isolated from Bothrops moojeni snake venom, towards leukemic cells J. Venom. Anim. Toxins incl. Trop. Dis.
Benati,Rogério Bodini; Costa,Tássia Rafaela; Cacemiro,Maira da Costa; Sampaio,Suely Vilela; Castro,Fabíola Attié de; Burin,Sandra Mara.
Abstract Background: Chronic myeloid leukemia (CML) is a BCR-ABL1+ myeloproliferative neoplasm marked by increased myeloproliferation and presence of leukemic cells resistant to apoptosis. The current first-line therapy for CML is administration of the tyrosine kinase inhibitors imatinib mesylate, dasatinib or nilotinib. Although effective to treat CML, some patients have become resistant to this therapy, leading to disease progression and death. Thus, the discovery of new compounds to improve CML therapy is still challenging. Here we addressed whether MjTX-I, a phospholipase A2 isolated from Bothrops moojeni snake venom, affects the viability of imatinib mesylate-resistant Bcr-Abl+ cell lines. Methods: We examined the cytotoxic and pro-apoptotic effect...
